Abstract

Objectives. This study aims to expand the knowledge on the hypoxia-activated signaling pathways that regulate proangiogenic events by using ameloblastoma cell lines in an in vitro study and will verify these findings with those obtained from a series of ameloblastoma cases from patients.
Methods. The project comprises two steps. In Step 1, the experimental design involves characterizing human ameloblastoma cells (HAM-2 and HAM-3) in vitro and exposing them to hypoxic conditions. The biological behavior of the cells, including proliferation, migration, and invasion, will be evaluated under hypoxia. Additionally, the expression of proangiogenic factors in these cells will be investigated, comparing hypoxic conditions with normal conditions. Hypoxia will be analyzed using a detection kit, and dynamic processes in cellular organization will be studied using high-resolution microscopy. Furthermore, immunocytochemistry will be performed to analyze the expression of proangiogenic factors. In Step 2, the findings will be tested using a series of ameloblastoma cases through immunohistochemistry compared with dental follicles. These results will be evaluated using machine learning techniques.
Expected results. This study will contribute to the understanding of the underlying mechanisms of tumor progression in ameloblastomas and may pave the way for the development of therapies specifically targeted at this type of tumor. The results obtained will help identify potential therapeutic targets and develop
more effective treatment strategies.
